Two-time premiership player David King said that Mifsud has lost the trust of the people he represented - the game's Indigenous players - and his job was untenable.
"The only honourable thing for him to do his to walk from the job," King told Fox Footy's AFL 360.
"Having made a couple of errors in the recent months or so, he has to walk.
"There is no other way this could play out."
The controversial comments made by Rendell - that clubs would only consider drafting Indigenous players if they had at least one white parent - were made in a private conversation with Mifsud.
Former Indigenous AFL player Dean Rioli says the Rendell and Neeld incidents proves that Mifsud can't be trusted to keep private conversations behind closed doors.
Rioli, who played 100 games for Essendon, echoed King's comments that the game's Indigenous community had lost faith in Mifsud.
"Everyone I speak to (say) he's very untrustworthy," Rioli told Melbourne radio station SEN.
"If you've got someone that's untrustworthy, you can't resolve anything privately and then you've got a serious issue.
"If he's going to run to the media every single time that someone comes to him with a question ... I wouldn't go and speak with him because I would be afraid that it would go straight to the headlines."
